Abstract
Objective To investigate the correlation between serum levels of long non-coding RNA small nucleolar RNA host gene 16(LncRNA SNHG16)and microRNA-106a-5p(miR-106a-5p)and the risk of early recurrence in patients with acute non-disabling ischemic stroke.Methods A total of 90 patients with acute non-disabling ischemic stroke who were admitted to Xi'an Qinhuang Hospital from March 2022 to December 2022 were included as the observation group,and 57 healthy individuals undergoing physical examinations during the same period were included as the healthy group.Based on the recurrence of acute non-disabling ischemic stroke within 1 year after discharge,the observation group was further divided into a recurrence group(31 cases)and a non-recurrence group(59 cases).The serum levels of LncRNA SNHG16 and miR-106a-5p were detected by RT-qPCR in both the observation and healthy groups and compared.Clinical data of the patients in the arecurrence and non-recurrence groups was collected and compared.ENCORI website was applied to predict the targeting relationship between LncRNA SNHG16 and miR-106a-5p.Pearson method was applied to analyze the correlation between LncRNA SNHG16 and miR-106a-5p.Multivariate Logistic regression analysis was used to identify the influence factors for the recurrence of acute non-disabling ischemic stroke,and receiver operating characteristic(ROC)curve analysis was applied to analyze the predictive value of serum LncRNA SNHG16 and miR-106a-5p for the early recurrence of acute non-disabling ischemic stroke.Results The serum level of LncRNA SNHG16 was lower in the observation group than that in the healthy group,while the level of miR-106a-5p was higher(t=15.129,9.328;P<0.01).The recurrence group had lower level of LncRNA SNHG16 and higher levels of miR-106a-5p,National Institutes of Health Stroke Scale(NIHSS)score,C-reactive protein(CRP),tumor necrosis factor-α(TNF-α),interleukin 6(IL-6),and vascular cell adhesion molecule-1(VCAM-1)compared with the non-recurrence group(t=8.453,8.951,19.690,19.233,7.435,4.812,3.352;P<0.05).LncRNA SNHG16 and miR-106a-5p had a targeting binding site,and they were negatively correlated(r=-0.483,P<0.05).NIHSS score,CRP,TNF-a,IL-6,VCAM-1,and miR-106a-5p were identified as risk factors for early recurrence of acute non-disabling ischemic stroke,while LncRNA SNHG16 was a protective factor(OR=1.523,1.359,1.632,1.533,1.646,1.728,0.764;P<0.05).The AUC values for predicting early recurrence of acute non-disabling ischemic stroke by LncRNA SNHG16 and miR-106a-5p were 0.934 and 0.901,respectively,and the combined prediction had an ACU of 0.978,indicating higher predictive value(Z=2.232,2.394;P<0.05).Conclusion Serum LncRNA SNHG16 and miR-106a-5p are influencing factors for the early recurrence in patients with acute non-disabling ischemic stroke and have a high predictive value for early recurrence.关键词
