Abstract
Objective:To establish a pre-column derivatization and high performance liquid phase method for the determination of total alkaloids in mulberry plants.Methods:Samples were collected with water and injected with 9-chloroformate methyl fluorene derivative reagent by HPLC with the volume of 20 µL.The chromatographic analysis was performed at 32℃with red spectrum RDC18(5 μm,4.6 x 250 mm)on the column and gradient elution of(citric acid buffer pH 4.2)(A)-acetonitrile(B)(75%B→47.5%B at 0~22 min,47.5%B→37.5%B at 22~22.01 min,37.5%B→37.5%B at 22.01~38 min,37.5%B→75%B at 38~38.01 min,75%B→75%B at 38.01~43 min).The flow rate was 1.5 mL/min.Results:In the linear range of 5~500 ug/mL,the linear relationship was good(r≥0.99),and the detection limit was 6.28 mg.kg-1.Conclusion:The method is rapid,simple and sensitive,and can be used for the determination of total alkaloid content in mulberry branch.关键词
