黄连制吴茱萸炮制前后抗炎、抗氧化、抑菌作用研究
Anti-Inflammatory,Anti-Oxidation,and Anti-Bacterial Effects of Evodiae Fructus Before and After Processing with Coptidis Rhizoma

Abstract
Objective To investigate the anti-inflammatory,anti-oxidation and anti-bacterial effects of Evodiae Fructus before and after processing with Coptidis Rhizoma.Methods Products of Evodiae Fructus(EF)and Evodiae Fructus processed with Coptidis Rhizoma(EFCR)were prepared.The anti-inflammatory effect in vivo and its effect on organ indexes were evaluated by the xylene-induced ear edema model in mice.A total of 66 KM mice were randomly divided into the blank control group(group A,equal volume of normal saline),the positive drug control group(group B,5 mg/kg dexamethasone acetate),the EF high-and low-dose groups(groups C1,C2;5,1 g/kg),and the EFCR high-dose and low-dose group(groups D1,D2;5,1 g/kg),with 11 mice in each group.The mice in each group were orally administered with the corresponding drug or 20 mL/kg normal saline once a day for 5 d.Lipopolysaccharide(LPS)was used to induce inflammatory reaction in mice mononuclear macrophages(RAW 264.7),and the effects of the blank control(group G,inoculated cells,containing equal volume of DMEM culture medium)and different mass concentrations(4,2,1,0.5,0.25 mg/mL)of EF(groups E1,E2,E3,E4,E5)and EFCR(groups F1,F2,F3,F4,F5)on cell survival rate were detected.The levels of nitric oxide(NO),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)in the blank group(group H,1 mL of DMEM culture medium),model group(group I,1 mL of DMEM culture medium containing 1 µg/mL LPS),EF high-,medium-,and low-dose groups(groups J1,J2,J3;1 mL of DMEM culture medium containing 1,0.5,0.25 mg/mL liquid medicine,and 1 µg/mL LPS),and EFCR high-,medium-,and low-dose groups(groups K1,K2,K3;1 mL DMEM culture medium containing 1,0.5,0.25 mg/mL liquid medicine,and 1 µg/mL LPS)were compared.The in vitro anti-oxidant activity was evaluated by 1,1-diphenyl-2-picrylhydazy(DPPH),and the in vitro anti-bacterial activity against Staphylococcus aureus and Escherichia coli was evaluated by the disk diffusion method and micro double dilution method.Results Compared with those in group A,the ear swelling degree of mice in groups B,C1 and C2 significantly decreased(P<0.05),the thymus index the spleen index of mice in groups B,D1,D2 significantly decreased(P<0.01),the spleen index of mice in groups B,C1,D1,and D2 significantly decreased(P<0.05),and the above indexes in groups D1 and D2 were significantly lower than those in groups C1 and C2,respectively(P<0.05).Compared with those in group G,the absorbance values in groups E1 and F1 significantly decreased(P<0.01),while the absorbance value in group E5 significantly increased(P<0.05).Compared with those in group I,the levels of NO in groups J1,J2,J3,K1,K2,and K3 significantly decreased(P<0.01);the levels of IL-6 in groups J1,J3,K1,and K3 significantly decreased(P<0.05),while the levels of TNF-α in groups J1 and K1 significantly decreased(P<0.05).Compared with those in group J1,the level of IL-6 in group K1 significantly decreased(P<0.05).Both EF and EFCR had a scavenging effect on DPPH free radicals,with half maximal inhibitory concentrations(IC50)of 0.074 3 mg/mL and 0.106 7 mg/mL,respectively.Both EF and EFCR were highly sensitive to the inhibition of Staphylococcus aureus,with inhibition zone diameters of(22.83±1.88)mm and(25.00±2.48)mm,respectively.However,they were less sensitive to the inhibition of Escherichia coli,with inhibition zone diameters of(7.83±0.85)mm and(7.17±0.94)mm,respectively.The minimum inhibitory concentration(MIC)of EF against Staphylococcus aureus was 15.63 mg/mL,which was lower than the 31.25 mg/mL of EFCR,while the minimum bactericidal concentration(MBC)of the two herbs was 62.5 mg/mL;the MIC and MBC for Escherichia coli were the same(62.5 mg/mL and 125.0 mg/mL,respectively).Conclusion Both EF and EFCR have anti-inflammatory,anti-oxidant,and anti-bacterial effects,but the latter has milder medicinal properties and stronger anti-inflammatory effect.关键词
