Effective Components and Mechanism of Dehumidification and Detoxification Formula in the Treatment of Eczema

Abstract
Objective To investigate the effective components and mechanism of Dehumidification and Detoxification Formula in the treatment of eczema.Methods The active components of Dehumidification and Detoxification Formula were screened by the TCMSP database,the active component-related targets were screened by the DrugBank database,eczema-related targets were screened by the GeneCards database,the active components of Dehumidification and Detoxification Formula and common targets of eczema were screened by the Python 3.8 programming software,the drug-component-disease target network was constructed by the Cytoscape 3.6.0 software,the protein-protein interaction(PPI)analysis was performed by the String database,and the Gene Ontology(GO)functional analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed by the DAVID database.A total of 50 BALB/C mice were randomly divided into the blank control group(group A),the model control group(group B),the positive drug control group(group C),and the Dehumidification and Detoxification Formula low-and high-dose groups(groups D1 and D2),with 10 mice in each group.The 5%2,4-dinitrochlorobenzene(DNCB)was applied to the abdomen of mice for sensitization twice and 1%DNCB was applied for stimulation four times to replicate the eczema model mice.After successful modeling,0.1 g of Dexamethasone Acetate Cream(30 g∶22.5 mg)was applied externally to the ears of mice in group C,and 5 mL of Dehumidification and Detoxification Formula(45 mg/mL and 180 mg/mL)were applied externally to the ears of mice in groups D1 and D2,respectively.Mice in groups A and B did not receive any special treatment.All mice were continuously administered for 12 d.The mass of mouse ear was weighted,and the morphology of ear tissue was observed by the hematoxylin-eosin(HE)staining.Enzyme-linked immunosorbent assay(ELISA)was used to detect the expression levels of serum tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),and interleukin-2(IL-2).Immunohistochemistry was used to detect the expression levels of TNF-α,IL-6,and IL-2 protein in mouse ear tissue.Results A total of 144 active components were screened,with 41 common targets.The core active components included quercetin,luteolin,kaempferol,baicalein,etc.The core targets included β2 adrenergic receptor(ADRB2),serine threonine protein kinase 1(Akt1),nitric oxide synthase(NOS2),TNF,IL-6,IL-2,etc.,involving signaling pathways such as TNF,IL-2,and IL-17.Compared with those in group B,the inflammatory cell infiltration and edema in the ears of mice in groups D1 and D2 reduced,and the pathological changes in the skin of mice in groups D1 and D2 improved;the mass difference in ear and the expression levels of serum TNF-α,IL-6,and IL-2 of the right ear tissue of mice in groups D1 and D2 significantly reduced(P<0.05),and the improvement effect of the above indicators was better in the group D2;the expression levels of TNF-α,IL-6,and IL-2 protein of the right ear tissue of mice in group D2 significantly reduced(P<0.05).Conclusion Network pharmacology and animal experiments can verify the effective components and mechanism of Dehumidification and Detoxification Formula in the treatment of eczema,which can provide a reference for its clinical application.关键词
