Effects of arid environment on the protein expression of lung in rats based on proteomics

Abstract

Objective:To screen differential expression proteins of rat lung tissue in arid environment and to explore the research targets of'dryness tends to impair the lung'and the potential mechanism of body acclimatization to arid environments.Methods:Twenty SD rats were randomly divided into normal control group and arid group,raised in SPF environment and simulated by arid environment in an artificial climate chamber respectively.The routine samples collection was conducted at the end of intervention.Lung tissue differential expression proteins of arid group rats were screened via proteomics techniques,candidate target verification were carried out via Western Blot and immunohistochemistry,and lung tissue morphology was observed via HE staining.Results:Significant differential protein grouping patterns between groups were revealed via Hierarchical Clustering analysis;882 kinds of differentially expressed proteins were mainly related to signal transduction mechanisms,protein post-translational modifications,and other sub-cellular structures such as cytoplasm,nucleus,and the scereted.They were mainly enriched in biological processes such as cellular metabolism,organelles,intracellular organelles,and other cellular components,and closely related to molecular functions such as protein binding and catalytic,and to adherens junction,and focal adhesion pathways.In Protein-Protein Interaction Network analysis,TRAF3,Myh10,and others were more critical in the lung tissue of drought treated rats.Target validation showed expression levels of TRAF3 and SP-A were up(P<0.01,P<0.05),XPR1 and SP-D were down(P<0.01,P<0.05).Conclusion:Long-term drought stress can induce alterations in protein expression within the lung tissue of healthy rats.The changes in the expression of SP-A,SP-D,TRAF3,and XPR1 constitute the molecular basis for'dryness evil tends to impair the lung'or the body's arid environment acclimatization.
