基于代谢组学探讨助孕宁Ⅱ号方对多囊卵巢综合征大鼠早期流产的作用机制
Exploration on the therapeutic mechanism of Zhuyunning Ⅱ Recipe on early abortion in polycystic ovary syndrome rats based on metabolomics

Abstract
Objective:To explore the blood metabolite profile,biomarkers and metabolic pathways of polycystic ovary syndrome(PCOS)threatened abortion based on metabolomics technology and the intervention effect of Zhuyunning Ⅱ Recipe.Methods:Thirty-six female SPF grade SD rats were selected and divided into 6 groups,including high-dose,medium-dose and low-dose of Zhuyunning Ⅱ Recipe groups,western medicine group,model group and blank group,with 6 rats in each group.Except the blank group,PCOS model rats were constructed by insulin and human chorionic gonadotropin(HCG)combined injection.Then letrozole was used to induce ovulation to construct PCOS threated abortion model.Starting from the first day of pregnancy,each group of rats was continuously administered with different drugs.On the seventh day of pregnancy,the rats were euthanized and their serum was collected.The metabolic profiles,biomarkers,and metabolic pathways of the six groups of serum were analyzed using ultra-high performance liquid chromatography-mass spectrometry.Results:The metabolic profile of PCOS threatened miscarriage rats showed significant positive and negative ion separation compared to the blank group.There were 33 potential biomarkers with abnormal expression in the blood of model group,which were mainly derived from phospholipid metabolism pathway,linoleic acid metabolism pathway and sex steroid hormone metabolism pathway.After treatment with three different doses of Zhuyunning Ⅱ Recipe,22 biomarkers were adjusted back.Conclusion:Glycerophospholipid metabolism,linoleic acid metabolism,sex steroid hormone production and metabolic pathway in PCOS threatened abortion rats were significantly changed,and the level of multiple biomarkers and metabolic pathways in PCOS threatened abortion rats could be regulated by Zhuyunning Ⅱ Recipe to improve the metabolism of sex hormone and glucose and lipid in vivo.关键词
