电针干预对高脂血症大鼠脂代谢及腺苷酸活化蛋白激酶信号通路的影响
Effects of electroacupuncture intervention on lipid metabolism and AMPK signal pathway in hyperlipidemia rats

Abstract
Objective:To observe the effects of electroacupuncture(EA)on gene and protein expression in hyperlipidemia rats,and to explore the possible mechanism of EA affecting AMPK pathway through adipocytokines to increase energy metabolism in vivo.Methods:Ninety male SD rats were randomly divided into 3 groups:high-fat diet and EA(HFD+EA)group,HFD group,normal diet(ND)group.HFD+EA group and HFD group were fed with high-fat diet for 8 weeks.HFD+EA group was intervened by EA at Zusanli(ST36)and Neiting(ST44)for 30 days after successful modeling.The lipid values in serum were detected by HITAI 7180 automatic biochemistry analyzer.Determinations of adiponectin and leptin in plasma were performed by ELISA.Adipose tissue and liver tissue were stained with HE.Protein expression was detected by Western Blot,and gene expression was detected by real-time fluorescence quantitative PCR.Results:Compared with HFD group,TG,TC and LDL-C levels in serum of rats in HFD+EA group decreased significantly,HDL-C level increased significantly(P<0.05),and the percentage of fat infiltration in liver tissue and the average cell area of adipose tissue were decreased significantly(P<0.05),the level of adiponectin was significantly increased(P<0.05),while the level of leptin was significantly decreased(P<0.05).The expression of pAMPK protein in hypothalamus,muscle and liver tissues increased significantly(P<0.05),AMPK-α2 mRNA in liver and muscle tissues and AMPK-α1 mRNA in hypothalamus tissues increased significantly(P<0.05).Conclusion:EA intervention significantly improves the blood lipid and tissue state of hyperlipidemia rats,and its mechanism may be related to regulating the content of adipocytokines and then activating AMPK signaling pathway.关键词
