Abstract

OBJECTIVE To investigate the protective effects of ultrasound-atomized Zanthoxylum bungeanum on allergic conjunctivitis(AC)in mice.METHODS An AC mouse model was established using ovalbumin(OVA).A total of 30 successfully modeled mice were randomly divided into a model group(MG),an olopatadine group(OH),a low-dose Zanthoxylum bungeanum group(PZ-L),a medium-dose Zanthoxylum bungeanum group(PZ-M),and a high-dose Zanthoxylum bungeanum group(PZ-H),with six mice in each group.A normal control group(CG)was also included.The PZ-L,PZ-M,and PZ-H groups received ultrasound-atomized decoctions of Zanthoxylum bungeanum at concentrations of 0.1,0.2,and 0.4 g/mL,respectively.The CG and MG groups received an equal duration of ultrasound-atomized 0.9%sodium chloride solution,while the OH group received olopatadine eye drops.Each mouse underwent atomization for 20 minutes daily for one week.After the final intervention,ocular signs were observed and scored,and serum and conjunctival tissues were collected.Hematoxylin-eosin(HE)staining was used to assess conjunctival structure,toluidine blue staining was employed to observe mast cell degranulation,and immunohistochemistry was used to detect immunoglobulin E(IgE)expression in the conjunctiva.Quantitative real-time PCR(RT-qPCR)was performed to measure mRNA expression levels of interleukin-4(IL-4)and tumor necrosis factor-α(TNF-α)in the conjunctiva,while enzyme-linked immunosorbent assay(ELISA)was used to quantify serum levels of substance P(SP),prostaglandin D2(PGD2),histamine(HIS),and TNF-α.RESULTS(1)Ocular sign scores:MG had significantly higher ocular sign scores than CG(t=15.543,P=0.000),while OH,PZ-L,PZ-M,and PZ-H groups had significantly lower scores than MG(all P<0.05).PZ-L and PZ-M groups had significantly higher scores than OH(all P<0.05).(2)Conjunctival structure:MG exhibited loose conjunctival structure,marked congestion and edema,substantial inflammatory cell infiltration in the lamina propria,and significant mast cell degranulation.These pathological changes were alleviated to varying degrees in OH,PZ-L,PZ-M,and PZ-H groups.(3)IgE expression:MG had significantly higher conjunctival IgE-positive expression rate than CG(t=18.078,P=0.000),while OH,PZ-L,PZ-M,and PZ-H groups had significantly lower rate than MG(all P<0.05).PZ-L and PZ-M groups had significantly higher rate than OH(all P<0.05).(4)IL-4 and TNF-α mRNA expression:MG had significantly higher conjunctival IL-4 and TNF-α mRNA expression than CG(both P<0.05).OH,PZ-L,PZ-M,and PZ-H groups exhibited significantly lower IL-4 and TNF-α mRNA expression than MG(all P<0.05).PZ-L and PZ-M groups had significantly higher IL-4 and TNF-α mRNA expression than OH(all P<0.05).(5)Inflammatory cytokines:MG had significantly higher serum SP,PGD2,HIS,and TNF-α levels than CG(all P<0.05).OH,PZ-L,PZ-M,and PZ-H groups exhibited significantly lower serum SP,PGD2,HIS,and TNF-α levels than MG(all P<0.05).PZ-L,PZ-M,and PZ-H groups had significantly higher serum SP,PGD2,HIS,and TNF-α levels than OH(all P<0.05).CONCLUSIONS Ultrasound-atomized Zanthoxylum bungeanum effectively alleviates allergic ocular symptoms in AC mice by modulating inflammatory pathways and reducing IgE protein levels.
