基于网络药理学和分子对接技术探讨黄芪消渴方治疗2型糖尿病作用机制
Exploration on the Mechanism of Huangqi Xiaoke Decoction in the Treatment of Type 2 Diabetes Mellitus Based on Network Pharmacology and Molecular Docking

Abstract
Objective To explore the mechanism of Huangqi Xiaoke Decoction in the treatment for type 2 diabetes mellitus(T2DM)by using network pharmacology and molecular docking.Methods TCMSP and SymMap databases were retrieved to obtain the active components of Huangqi Xiaoke Decoction and potential targets were predicted.GeneCards and DisGeNET databases were used to obtain the related targets of T2DM.A Venn diagram was drawn to identify common targets between the active components and the disease.The Cytoscape 3.7.2 software was used to construct the active component-target network,PPI network and core target network.GO and KEGG pathway enrichment analysis were performed using Metascape.Finally,AutoDockTools and PyMOL software were used for molecular docking verification.Results Totally 142 active components and 2 458 disease targets of Huangqi Xiaoke Decoction were included,with 437 potential targets for treating T2DM,including AKT1,GAPDH,IL6,etc.;a total of 3 802 GO functional terms and 248 KEGG pathways were enriched.Molecular docking results indicated that the components binds stably to the core target proteins.Conclusion Huangqi Xiaoke Decoction may exert its therapeutic effects on T2DM through targets such as AKT1,GAPDH and IL6,as well as signaling pathways including cancer pathways,insulin resistance and calcium signaling pathways.关键词
