Correlation of Peripheral Th9 Cells and its Cytokines with the IBS-SSS Score in Patients with Irritable Bowel Syndrome

Abstract
Objective To investigate the correlation of the proportion of T helper 9(Th9)cells and the expression level of interleukin-9(IL-9)in peripheral blood with the Irritable Bowel Syndrome Symptom Severity Scale(IBS-SSS)score in patients with irritable bowel syndrome(IBS).Methods A total of 103 IBS patients treated in the Department of Gastroenterology,Hengshui People's Hospital from January 2022 to June 2024 were enrolled in the study group,and 103 healthy people were enrolled in the control group.The proportion of Th9 cells and the expression level of IL-9 in peripheral blood were detected by flow cytometry and enzyme-linked immunosorbent assay(ELISA),respectively.The IBS severity was assessed by the IBS-SSS.The correlation of Th9 cell proportion,and IL-9 expression level with the IBS-SSS score was analyzed by Spearman correlation analysis.Factors influencing the occurrence of IBS were defined by multivariate Logistic regression analysis.Receiver operating curve(ROC)analysis was used to evaluate the value of Th9 cell proportion and IL-9 expression level in diagnosing IBS.Results The proportion of Th9 cells and serum IL-9 in the study group were significantly higher than those of the control group(P<0.05).Th9 cell proportion,IL-9 expression level and IBS-SSS score in the moderate-to-severe group were significantly higher than those of the mild group(P<0.05).The IBS-SSS score was positively correlated with the proportion of Th9 cells and the expression level of IL-9(P<0.05).Multivariate Logistic regression analysis showed that the high proportion of Th9 cells and the high level of IL-9 were both risk factors for IBS.ROC curve analysis showed that the area under the curve(AUC)of Th9 cell proportion and IL-9 expression level in diagnosing IBS was 0.699(95%CI:0.584-0.813)and 0.707(95%CI:0.606-0.808),respectively,and the AUC of their combination was 0.851(95%CI:0.768-0.933).Conclusion The proportion of Th9 cells and the expression level of IL-9 in peripheral blood of IBS patients are elevated,and they are positively correlated with the disease severity.The proportion of Th9 cells and the expression level of IL-9 have good diagnostic abilities on IBS.关键词
