The Role of TGF-β1/SMAD3 Signaling Pathway in Back Meridian Tuina Promoting NK Cell Activity in Chronic Fatigue Syndrome Rats

Abstract
Objective:To investigate the effects of back meridian tuina on the behavior,natural killer(NK)cell activity,and the TGF-β1/SMAD3 signaling pathway in rats with chronic fatigue syndrome(CFS).Methods:Twenty-four male Sprague-Dawley rats were randomly divided into four groups:control group,model group,back meridian tuina group,and ginsenoside group,with six rats in each group.The CFS rat model was established using chronic fatigue dual stress method(chronic restraint plus forced swimming in cold water).Back meridian tuina group received a sequence of treatments including flat pushing along the"Governor Vessel and Bladder Meridian"for 4 min,thumb pressing and kneading on"Back-shu acupoints"for 4 min,and finger pressing on"Back-shu acupoints"for 4 min,at a rate of 70~90 times/min,for 12 min per session,over 4 consecutive weeks.The general condition of the rats was assessed using a semi-quantitative scoring system.The open field test was used to evaluate anxiety and depression-like be-haviors,and the exhaustive swimming time was measured to assess fatigue.NK cell activity in the spleen tissue was detected using the LDH release method.Serum levels of transforming growth factor-beta1(TGF-β1)and interferon-gamma(IFN-γ)were measured using ELISA.Real-time quantitative PCR was used to determine the mRNA expression levels of TGF-β receptor 1(TβR1)and SMAD3 in spleen tissue.Results:Compared with the control group,the model group showed significantly increased semi-quantitative scores for general condition(P<0.05),significantly reduced open field activity time and exhaustive swimming time(P<0.05),significantly decreased NK cell activity in spleen tissue(P<0.05),and significantly increased mRNA expression of TβR1 and SMAD3(P<0.05).Serum TGF-β1 levels were significantly increased(P<0.05),while IFN-γ levels were significantly decreased(P<0.05).Compared with the model group,the back meridian tuina group showed significantly decreased semi-quantitative scores for general condition(P<0.05),significantly increased open field activity time and exhaustive swimming time(P<0.05),increased NK cell activity(P<0.05),and significantly decreased mRNA expression of TβR1 and SMAD3(P<0.05).Serum TGF-β1 levels were sig-nificantly decreased(P<0.05),while IFN-γ levels were significantly increased(P<0.05).Conclusion:Back meridian tuina can effec-tively alleviate fatigue and other symptoms in CFS rats,restoring NK cell activity.This effect may be related to the regulation of the TGF-β1/SMAD3 signaling pathway in NK cells and the secretion of related immune factors.关键词
