摘要

Abstract

Objective To study the effecacy and mechanism of moxibustion on stress urinary incontinence(SUI)in rats.Methods SUI rat model was established by bilateral ovariectomy combined with vaginal dilatation.Forty SD rats were randomly divided into 3 groups according to the random number table:The moxibustion intervention group(Mox)(n=15)was treated with moxibustion,the control group(NC)(n=15)was only treated with bilateral oophorectomy combined with vaginal dilatation,and the blank group(n=10)did not undergo any interventions.The leak point pressure(LPP)was measured,and Sirius red staining of urethral tissue and Van Gieson staining(EVG),real-time fluorescent quantitative PCR(qRT-PCR)were used to detect the mRNA levels of collagen Ⅰ,Ⅲand α-actin(α-SAM),and Western blot was used to detect the expression of TGF-β1 and CTGF to evaluate the therapeutic effect and molecular mechanism of moxibustion on urinary incontinence.Results Compared with the blank group,the LPP of the NC group and the Mox group decreased significantly(P<0.01),the Sirius red staining and EVG staining showed that the positive area of collagen and elastic fibers decreased(P<0.01),and the mRNA levels of collagen Ⅰ,collagen Ⅲ and α-SAM decreased significantly(P<0.01).The protein expression of TGF-β1 and CTGF was down-regulated.Compared with the control group,the LPP,the proportion of collagen and elastic fibers in the urethral tissue,the mRNA levels of collagen Ⅰ,collagen Ⅲ and α-SAM,and the protein expression of TGF-β1 and CTGF were significantly increased in the Mox group(P<0.01).Conclusion Moxibustion can improve the urination function of SUI rats and reduce urethral injury,which may relieve urinary incontinence by activating TGF-β1/CTGF pathway to restore collagen and elastic fiber structure in urethral tissue.
