温肾活血解郁方对卵巢去除诱导的骨质疏松大鼠PI3K/AKT/mTOR通路表达调节作用研究
Study on the regulatory effect of Wenshen Huoxue Jieyu Formula on the expression of PI3K/AKT/mTOR pathway in osteoporosis rats induced by ovarian removal

Abstract
Objective To explore the regulatory effect of Wenshen Huoxue Jieyu Formula on PI3K/AKT/mTOR pathway expression in osteoporosis rats.Methods 40 SD rats were randomly divided into four groups,namely the sham operation group,the model group,the treatment group(treated with Wenshen Huoxue Jieyu Formula)and the treatment group+LY294002 group,with 10 rats in each group.Except for the sham operation group,which was only ovariectomized and exposed without removing the ovaries,the ovaries were ligated and removed in the other three groups.After 12 weeks of treatment with Wenshen Huoxue Jieyu Formula,bone mineral density(BMD),bone trabecular thickness(Tb.Tn),bone volume/total volume(BV/TV),trabeculae number(Tb.N),and trabeculae separation(Tb.Sp)were evaluated in each group;The serum levels of alkaline phosphatase(ALP),osteocalcin(OC),and C-terminal telopeptide(CTX)were detected in rats by enzyme-linked immunosorbent assay(ELISA);real-time quantitative reverse transcription PCR(qRT-PCR)was used to detect the levels of Runt-related transcription factor 2(Runx2),collagen typeⅠ alpha 1(CoL1A1),nuclear factor κB ligand receptor activator(RANKL),Osterix,and osteoprotegerin(OPG);And Western blot was used to detect the expression of PI3K/AKT/mTOR pathway proteins.Results Compared with the sham operation group,BMD,BV/TV,Tb.N,Tb.Th,ALP,OC,Runx2,Osterix,CoL1A1,OPG,p-PI3K/PI3K,p-AKT/AKT,and p-mTOR/mTOR were significantly decreased in the model group,while Tb.Sp,CTX,and RANKL were significantly elevated(P<0.05);Compared with the model group,BMD,BV/TV,Tb.N,Tb.Th,ALP,OC,Runx2,Osterix,CoL1A1,OPG,p-PI3K/PI3K,p-AKT/AKT,and p-mTOR/mTOR were significantly increased in the treatment group after treatment with the Wenshen Huoxue Jieyu Formula,while Tb.Sp,CTX,and RANKL were decreased(P<0.05);Compared with the treatment group,BMD,BV/TV,Tb.N,Tb.Th,ALP,OC,Runx2,Osterix,CoL1A1,OPG,p-PI3K/PI3K,p-AKT/AKT,and p-mTOR/mTOR were significantly reduced in the treatment group+LY294002 group after injection of LY294002,while Tb.Sp,CTX,and RANKL were significantly elevated(P<0.05).Conclusion By activating PI3K/AKT/mTOR signaling pathway,Wenshen Huoxue Jieyu Formula plays a role in bone protection,thereby alleviating osteoporosis induced by ovarian removal.关键词
