摘要

Abstract

Objective To investigate the mechanism of action of Scutellaria barbata polysaccharide nano selenium in treating inflammation in liver cancer model mice.Methods 24 male BALB/c mice were selected and randomly divided into four groups,with 6 mice in each group.Three groups were set up as the experiment groups,and HepG2 tumor bearing mouse models were established by subcutaneous injection of HepG2 liver cancer cells.They were named the model group,Scutellaria barbata polysaccharide group(SBP),and Scutellaria barbata polysaccharide nano selenium group(SBP SeNPs),respectively.The other group of unmodeled mice was the blank group.After 15 days of corresponding administration,the mice were euthanized.The blood was extracted from the eyeball,and the concentrations of serum interleukin(IL)-6,IL-1β,and tumor necrosis factor-α(TNF-α)were measured by electrochemiluminescence assay(ECLIA).The phagocytic ability of mouse peritoneal macrophages was measured;the spleen was extracted,a spleen cell suspension was prepared,and the proliferation ability of splenic lymphocytes was measured;liver tissue was extracted,proteins were extracted,and the expression levels of P-ERK/ERK and P-P38/P38 proteins were measured by Western blot.Results Compared with the blank group,the expression levels of inflammatory factors of IL-6,IL-1β,and TNF-α in the serum of the model group increased;The phagocytic ability of peritoneal macrophages and the proliferation ability of splenic lymphocytes in mice decreased;Western blot results showed an increase in the expression levels of P-ERK/ERK and P-P38/P38 proteins.Compared with the model group,the expression levels of inflammatory factors of IL-6,IL-1β,and TNF-α in the serum of the SBP group and SBP SeNPs group were significantly decreased,and the phagocytic ability of peritoneal macrophages and the proliferation ability of splenic lymphocytes in mice were significantly increased.Western blot results showed a decrease in the expression levels of P-ERK/ERK and P-P38/P38 proteins.Conclusion Scutellaria barbata polysaccharide nano selenium can effectively improve the inflammatory response of liver cancer model mice,promote the increase of macrophage phagocytic ability and lymphocyte proliferation,and its effect is superior to that of Scutellaria barbata polysaccharide.Its mechanism of action may be the activation of immune regulation mechanism,which is achieved by regulating the ERK/P38/MAPK signaling pathway and reducing the expression levels of inflammatory factors such as IL-6,IL-1β,TNF-α,etc.
