Analysis of platelet function changes during 4℃ refrigerated storage in two types of storage bags

Abstract
Objective To compare the platelet count and functional changes of leukocyte-depleted pooled plate-let concentrates stored at 4℃ in two different types of storage bags with those stored at 22℃ under continuous agitation.Methods Leukocyte-depleted pooled platelet concentrates were divided into three groups:Group A,stored at 4℃ in standard blood storage bags;Group B,stored at 4℃ in platelet storage bags;and the Control Group,stored at 22℃ with agitation in platelet storage bags.Samples were collected at days 1,3,5,7,10,15,and 21 for Groups A and B,and at days 1,3,5,and 7 for the Control Group.Platelet parameters were assessed using a hematology analyzer,thromboelas-tography(TEG),pH meter,and spectrophotometer.Results Both Groups A and B exhibited a gradual decline in plate-let count over the storage period,yet the total platelet count per bag remained above 2.5 × 1011 even at day 21.In con-trast,the Control Group maintained a relatively stable platelet count over 7 days(P>0.05).By days 15-21,throm-boelastography parameters(MA,R,K,α-angle)and lactate levels in Groups A and B were comparable to those in the Control Group at day 7(P>0.05).The final pH values of all three groups remained above 7.0 at the end of their respec-tive storage periods.Conclusion Compared to platelets stored at 22℃ with agitation,platelets stored at 4℃ under static conditions demonstrated superior aggregation activity and a lower metabolic rate.No significant differences were observed between standard blood storage bags and platelet storage bags when storing leukocyte-depleted pooled platelet concen-trates at 4℃.关键词
