海南砂仁化学成分及其胃黏膜保护活性研究
Study on the chemical composition of Amomum longiligulare T.L.Wu and its protective activity of gastric mucosa in vitro

Abstract
Objective:In this paper,the chemical constituents of Amomum longiligulare T.L.Wu were isolated and the in vi-tro gastric mucoprotective activity of its monomers was investigated.Methods:Amomum longiligulare T.L.Wu 70%alcoholic extract was extracted by systematic solvent extraction to obtain the dichloromethane part,which was isolated and purified by silica gel column chromatography,Sephadex LH-20 gel column chromatography,ODS column chromatography,semi-preparative high performance liquid chromatography,etc.,and the structures of the compounds were identified by modern wave spectrometry;the compounds obtained from the isolation were evaluated and detected by IL-1β,IL-6,and PEG2 factor expression by using the CCK-8 method and ELISA kits in vitro.The isolated compounds were evaluated by CCK-8 method and ELISA kit,and the ex-pressions of IL-1β,IL-6 and PEG2 factors were detected.Results:Sixteen compounds were isolated from the dichloromethane site of Amomum longiligulare T.L.Wu,including 11 linear diarylheptanes,three flavonoids,and two other classes,which were identified as 5-hydroxy-3,7,4′-trimethoxyflavone(1),hydroquinone(2),1,7-bis(4-hydroxyphenyl)-3-heptanone(3),5,4′-dihy-droxy-7-methoxyflavone(4),(3R,5R)-3-acetoxy-5-hydroxy-1,7-bis(3,4-dihydroxyphenyl)heptane(16),where compounds 1-6,10-14,and 16 turned out to be protective against ethanol-induced GES-1 cell damage.Compounds 11,13,and 14 decreased the expression of pro-inflammatory factors IL-1β and IL-6 and elevated the expression of gastric mucosal protective factor PEG2 in cell supernatants.Conclusion:Compounds 1-2,4-6,9,13-16 were isolated from Amomum longiligulare T.L.Wu for the first time,and compounds 1-6,10-14,16 have certain in vitro gastric mucoprotective activities,of which compounds 11,13,14 can reduce the expression of pro-inflammatory factors IL-1β,IL-6 and elevate the expression of gastric mucoprotective factor PEG2 in the supernatant of cells.关键词
