基于生物信息学和临床样本探究FMR1与胃癌的相关性及其在肿瘤微环境中的作用
Exploration of the perturbation of FMR1 in gastric cancer and the role in tumor microenvironment based on bioinformatics and clinical samples

Abstract
Objective:To analyze the expression differences of Fragile X Mental Retardation 1(FMR1)in gastric cancer and adjacent non-tumor tissues,investigate its correlation with the clinical pathological characteristics and outcomes of gastric cancer patients.And to analyze the potential signaling pathways regulated by FMR1 and its role in the tumor microenvironment.Methods:The TIMER2.0 tool was used to analyze FMR1 in the pan-cancer.The Cancer Genome Atlas(TCGA)database was used to ana-lyze differences in the expression of the FMR1 gene between gastric cancer tissues and adjacent normal tissues,and the results were verified by qRT-PCR and immunohistochemistry.The Kaplan-Meier Plottery database was used for survival analysis.R soft-ware and clinical samples were used to assess the correlation between FMR1 expression and clinical pathological factors in gastric cancer patients.The relationship between FMR1 and tumor immune infiltration in gastric cancer was analyzed through the TISIDB database.The function of FMR1 and its interacting genes was analyzed by functional annotation tools.Results:The positive ex-pression rate of FMR1 in gastric cancer was significantly higher than in normal gastric tissue(P<0.001).Patients with high FMR1 expression had a significantly reduced overall survival time(P<0.05).The varying expression of FMR1 in gastric cancer patients was associated with the activity of various immune cells in their tumor microenvironment(P<0.05).FMR1 may be in-volved in processes such as keratin filament and gated channel activities,and was associated with pathways related to olfactory sig-nal transduction,African sleeping sickness,and PPAR signaling.Bioinformatics and clinical sample analysis indicated that the ex-pression level of FMR1 was closely related to the staging of gastric cancer patients(P<0.05).Conclusion:The high expression level of the FMR1 gene is closely associated with poor prognosis and immune cell infiltration in gastric cancer patients,suggesting its potential to be established as a novel biomarker for the diagnosis and treatment of gastric cancer.关键词
