Effects of baicalin on viability,proliferation,apoptosis,and inflammation levels of human normal pancreatic ductal epithelial cells(HPDE6-C7)induced by LPS

Abstract
Objective To investigate the effects of baicalin on the viability,proliferation,apoptosis and inflammation of human normal pancreatic ductal epithelial cells(HPDE6-C7)induced by lipopolysaccharide(LPS).Methods The potential compound targets of baicalin were screened by Swiss TargetPrediction.The Comprehensive Human Gene Database(Genecard)was searched for disease targets related to pancreatitis.The disease targets were integrated with baicalin compound targets to construct a Venn diagram.The David Database was used to perform a gene ontology(GO)functional enrichment analysis and a Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis to clarify the mechanism of baicalin in the treatment of pancreatitis and the related pathways.The effects of different concentrations of baicalin on the viability,proliferation,apoptosis and inflammation of HPDE6-C7 cells induced by lipopolysaccharide(LPS)were detected by cell counting kit-8(CCK-8),5-acetyl-2-deoxyuridine(EdU),flow cytometry and Elisa inflammation kit.Western blot was used to detect the protein expression of BCL-2 and IκBα.Results Bioinformatics identified 100 targets of baicalin and 22620 targets of pancreatitis,and 92 intersections were taken.The David Database analysis showed that the KEGG of baicalin in pancreatitis was mainly in apoptosis,TNF signaling pathway and NF-κB signaling pathway,and played a molecular function of apoptosis in regulating the biological process of reactive oxygen species metabolism and mainly in the cell composition of the cell membrane.The results of in vitro cell experiments showed that compared with the control group,baicalin significantly alleviated the decrease of cell viability and proliferation(P<0.05),the increase of apoptosis level,the decrease of apoptosis marker protein BCL-2(P<0.05),and the increase of IL-15,IL-6 and TNF-6 levels(P<0.05).Western blot results showed that baicalin could regulate the activation of NF-κB signaling pathway in cells.Conclusion Baicalin can reverse the effects of LPS on the viability,proliferation,apoptosis and inflammation level of HPDE6-C7 cells.关键词
