Abstract

Cordyceps chanhua fruiting body is a traditional Chinese medicine with a long history of application.In the present study,the role of microRNA in the growth and development of spore(SP)and fruiting body(FB)of C.chanhua was investigated based on Small RNA sequencing method,and the structural features and expression of the obtained miRNAs were investigated by the use of bioinformatics software and validated by Stem-loop RT-PCR.In total,501 new predicted miRNAs were identified,with lengths ranging from 18 to 25 nucleotides(nt)and a U bias at the first base.There were 52 DEmiRNAs,of which 15 were up-regulated and 37 down-regulated.Two up-regulated and 2 down-regulated DEmiRNAs were randomly selected for Stem-loop RT-PCR validation.The results showed that all four DEmiRNAs were truly expressed.A total of 1 323 target genes was predicted for all DEmiRNAs,of which 470,568,673,1 068 and 388 target genes could be annotated to COG,KOG,eggNOG,Pfam and Swissprot databases,respectively.In addition,966 and 415 target genes could be annotated to 35 functional terms in GO and 50 metabolic pathways in KEGG database,respectively.This study reveals the presence and expression of miRNAs in C.chanhua,and these miRNAs are involved in the life activities of spores and fruiting bodies by regulating the expression of potential genes.
