金针菇野生菌株原生质体单核化和无性孢子单孢分离中出现的核偏分离现象

Abstract
Two nuclei in the same cell of basidiomycetous edible fungi have different activeness,which will lead to the phenomenon of nuclear partial segregation in protoplast mononucleation and asexual spore monosporous isolation.In this study,15 wild strains(GS0002,GS0049,GS0116,GS0145,GS0146,GS0169,GS0174,GS0175,GS0180,GS0193,GS0194,GS0196,GS0200,GS0216,GS0225)and 2 cultivated strains(T011,T022)of Flammulina filiformis were used as test materials to isolate the monocaryon by the method of protoplasmic mononucleation.The results showed that 13 wild strains and 2 cultivated strains showed partial segregation in the process of protoplast mononucleation,with mating type deviation ratios ranged from 1.9∶1 to 35∶0.Five wild strains were selected,to isolate the mononucleosomes by the method of single-spore isolation of oidium.The results showed that 4 wild strains(GS0116,GS0169,GS0200,GS0216)showed partial segregation during oidium single-spore isolation,with mating type deviation ratios ranged from 1.7∶1 to 26.5∶1.The strong and weak nuclei of the three strains that showed mating type partial segregation occurred in both isolation methods were identified by hybridization affinity experiments.As a result,their mating types were found to be consistent(GS116-6 and GS0116-107,GS0116-17 and GS0116-30,GS0169-2 and GS0169-121,GS0169-1 and GS0169-79,GS0200-30 and GS0200-1,GS0200-21 and GS0200-110).关键词
