Abstract

It had been reported that Mycena sp.MF23 promoted the growth of Dendrobium officinale.In present study,the molecular identification of MF23 was conducted for the first time,and the effects of co-culture between MF23 and D.officinale on biomass of the fungus and plant,and on physicochemical properties and plant-hormone content of the substrates were investigated.MF23 was identified as Mycena purpureofusca.After 4 months of growth,the dry weights of root,stem and leaf of D.officinale in co-cultured group(FP)were significantly increased by 18.5%-41.0%as compared with those in plant pure-cultured group(P).Meanwhile,the ergosterol content,and the laccase and lignin peroxidase activities of FP substrates were significantly increased by 11.8%,41.6%and 133.3%respectively as compared with those of the fungus pure-cultured group(F).At this time,there was no significant difference in nitrate nitrogen content between FP and F substrates;however,the content of ammonium and amino nitrogen of FP was significantly increased by 9.3%and 41.1%respectively as compared with that of F.There was no cis-zeatinriboside(cZR)in sterilized pine bark substrate(control)and substrates of P that grew for 4 months.At this time,the cZR content of F substrates was(6.37±0.79)ng/g,the cZR content of FP substrates was(0.66±0.08)ng/g;the indoleacetic acid(IAA)content was significantly increased by 28.1%as compared with that of CK(control),and the IAA content was significantly increased by 36.2%as compared with that of P.These results showed that MF23 could promote the growth of D.officinale by producing plant hormone.The co-culture system had higher nitrogen-utilization efficiency.This study provided references for the further mechanism researches on growth promoting fungi in orchids.
