Optimization of extraction conditions and antioxidant activities of polysaccharides from Inonotus hispidus fruiting bodies

Abstract
The optimal extraction conditions of polysaccharides from Inonotus hispidus fruiting bodies were obtained by optimizing the extraction process using single factor experiment combined with response surface methodology.The optimal extraction conditions were as follows:liquid-solid material ratio of 27.9∶1(mL/g),hot water extraction temperature of 86.2 ℃,and extraction time of 3.4 hours.Under these conditions,the extraction rate of polysaccharides was 6.815%.Determination of the radical scavenging activities of 1,1-diphenyl-2-picrylhydrazyl(DPPH)and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)of the obtained polysaccharides showed that at 10 mg/mL the clearance rates of DPPH and ABTS were 88.97%and 77.51%,respectively.The cognitive ability of D-galactose-induced mice was evaluated through open field experiments;The levels of superoxide dismutase(SOD),malondialdehyde(MDA),and reactive oxygen species(ROS)were detected by using reagent kits;Western blot was used to detect the expression of nuclear factor erythroid-2-related factor 2(Nrf2)and heme oxygenase-1(HO-1).Compared with the model group,the level of SOD increased(P<0.01),that of MDA and ROS decreased(P<0.01),and the expression levels of Nrf2 and HO-1 increased in the brains of mice treated with oral polysaccharides.In summary,in vitro and in vivo experimental results indicate that polysaccharides from I.hispidus fruiting bodies can effectively alleviate cognitive impairment induced by D-galactose in mice and alleviate oxidative stress in the brain through their antioxidant effects.This study provides reference material for further study of polysaccharides from I.hispidus fruiting bodies in the field of food and medicine.关键词
