Expression and significance of ENO2 in oral squamous cell carcinoma

Abstract
Objective:To evaluate the expression levels and clinical significance of enolase 2(ENO2)in oral squa-mous cell carcinoma(OSCC).Methods:Three gene expression profile datasets were obtained from the GEO database,and differentially expressed genes(DEGs)were identified.GO and KEGG pathway analyses were subsequently performed.Bioin-formatics methods were utilized to assess the expression level of ENO2.Immunohistochemistry(IHC)was employed to assess the expression levels of ENO2 in 106 cases of oral squamous cell carcinoma(OSCC)tissues and 45 cases of adjacent tissues.Additionally,Western blotting and quantitative PCR(qPCR)were utilized to evaluate the expression levels of ENO2 protein and mRNA in human oral keratinocyte(HOK)cells,SCC15 cells,and CAL27 cells.Results:Bioinformatics analysis revealed that ENO2 expression in OSCC tissues was elevated,and the overall survival time of OSCC patients with high ENO2 expres-sion was significantly shorter than that of patients with low expression(P<0.05).The results indicated that the ENO2 protein expression level in OSCC tissues was significantly higher compared to adjacent normal tissues(P<0.05).Furthermore,the expression level of ENO2 protein was significantly associated with the TNM stage,T stage,lymph node metastasis,and differ-entiation of OSCC patients.The expression levels of ENO2 mRNA and protein in OSCC cells were also higher than in oral ep-ithelial cells(P<0.05).Conclusion:ENO2 is significantly overexpressed in OSCC tissues and cells.Its expression is associ-ated with lymph node metastasis,tumor differentiation,and poor prognosis in OSCC patients,making it a potential prognostic biomarker.Furthermore,ENO2 may serve as a novel therapeutic target for OSCC.关键词
