摘要

Abstract

Objective:To investigate the short-term effects of fluoride varnish on the oral microbial diversity,commu-nity structure,and functional genes of preschool children with varying caries levels,thereby providing evidence for personal-ized caries prevention strategies.Methods:Sixty 36-month-old children were recruited and categorized into no caries,low caries,and high caries groups.Supragingival plaque samples were collected at baseline,Day 1,Day 3,and Day 7 post-fluoride application.Microbial diversity,community structure,and functional gene changes were analyzed using 16S rRNA high-throughput sequencing.Results:After fluoride application,the diversity of microbiomes significantly decreased in all groups(P<0.01),with the high-caries group exhibiting the greatest change and the slowest dynamic adjustment of microbiome di-versity and structure.Principal component analysis revealed that the community structure of the high-caries group deviated the farthest from baseline on the 1st day post-fluoride application(P<0.01),and on the 7th day,the diversity and structure of the microbiome had not fully returned to pre-application status(P<0.05).Functional prediction analysis indicated that the abun-dance of sugar metabolism-related genes in the high-caries group was significantly higher than in the other groups at baseline(P<0.001),which decreased significantly post-fluoride application(P<0.01),while the abundance of fluoride resistance genes was significantly upregulated(P<0.001).Conclusion:The short-term effects of fluoride varnish on oral microbial com-munities vary significantly by caries level,highlighting the necessity for personalized caries prevention strategies and guiding future multi-omics research.
