Study on the suspended cell culture-based Newcastle disease virus in BHK-21 and immunogenicity evaluation

Abstract
This study aimed to establish an efficient suspension culture process for Newcastle disease virus(NDV)strain A-Ⅶ in BHK-21 cells and evaluate its immunogenicity.Key parameters,including initial cell density,harvest time,virus-to-cell ratio,and cultivation temperature,were optimized to enhance viral production.Subsequently,the immunogenicity of the vaccine produced under optimized conditions was assessed.Results demonstrated that the highest viral yield was achieved under the following optimal conditions:initial BHK-21 cell density of 4.12×10⁶ cells/mL,virus-to-cell volume ratio of 0.02%,and harvest at 72 hours post-inoculation with cultivation at 35℃.The hemagglutination(HA)titer of the suspension-cultured virus reached 1:512.Comparative immunogenicity analysis in SPF chickens revealed that the hemagglutination inhibition(HI)antibody titers induced by the suspension-cultured virus were significantly lower than those elicited by the egg-cultured virus(P<0.05).These findings suggest that the suspension culture process holds significant potential for NDV strain A-Ⅶ production,but further optimization is required to improve its immunogenicity and enhance antibody response levels.关键词
