Abstract

The experiment aimed to investigate the effect of culture medium osmotic pressure on the expression of porcine epidemic diarrhea virus(PEDV).PEDV was inoculated into serum-free medium containing ST suspension cells,and sodium chloride was added to adjust the osmotic pressure of the cell culture medium during the viral expression phase.The effects of varying osmotic pressure conditions on cell growth,metabolic activity,and viral titer were evaluated.Results showed that maintaining the osmotic pressure of the culture medium at(360±10)mOsm/kg during viral expression by sodium chloride supplementation increased the viral titer from 7.50 Log TCID50/mL to 8.80 Log TCID50/mL,significantly enhancing the harvested PEDV antigen or viral seed titer.This study demonstrates that regulating the osmotic pressure of the culture medium can markedly improve PEDV viral titer during expression.
