Function study of VrJAZ1 and VrJAZ12 in the resistance of mung bean to Pythium myriotylum

Abstract
[Objectives]This study aimed to explore the function of JAZ(jasmonate ZIM-domain)genes in the resistance of mung beans to Pythium myriotylum,providing references for the control of root rot disease in mung beans.[Methods]The JAZ family in mung beans was identified and analyzed for its expression pattern following P.myriotylum infection.Using transcriptome data and real-time quantitative PCR(q-PCR),two target genes were selected.Vectors for overexpression and gene editing of these genes were constructed.The function of these genes in resistance to P.myriotylum was studied through transient transformation in tobacco and transgenic expression in mung bean hairy roots.Additionally,after spraying methyl jasmonate(MeJA)on'Sulü No.1'seedlings,changes in expression patterns were analyzed.[Results]P.myriotylum could infect mung bean leaves and the stem-root junction.From the mung bean JAZ gene family,VrJAZ1 and VrJAZ12 were identified as target genes whose expression levels decreased with prolonged infection time.After spraying MeJA,the expression patterns of VrJAZ1 and VrJAZ12 changed.Overexpressing these target genes decreased resistance to P.myriotylum,while gene editing of these genes increased resistance.[Conclusions]VrJAZ1 and VrJAZ12 negatively regulated the resistance of mung beans to P.myriotylum by modulating the JA signaling pathway.关键词
