Abstract
[Objectives]The aim of this study was to isolate and identify a novel bovine herpesvirus 4(BoHV-4)strain in China,associated with reproductive disorders in large-scale dairy herds.[Methods]Swabs were collected from the reproductive tracts of cattle suspected to be infected with BoHV-4 at a cattle farm in the Northeast China.Positive samples were inoculated with HeLa cells for virus isolation,followed by identification using specific qPCR,indirect immunofluorescence(IFA),and electron microscopy.One step growth curve of the virus was determined.The gB and TK genes of the virus were amplified through PCR and analyzed for genetic evolution.[Results]Positive samples were inoculated with cells.After three generations of transmission,the cells exhibited typical cytological effects(CPE)characteristic of herpesviruses such as round contraction and aggregation.IFA results revealed distinct green fluorescence specifically in infected cells.Electron microscopy images showed infected cells with a diameter ranging of 100-150 nm,displaying clear capsid and capsule structures.The isolated strain named TL01 demonstrated a growth curve peaking at 36 hours post-infection(106.3 TCID50·mL-1).PCR amplification confirmed that gB gene belonged to genotype 2 along with reference strain DN599 while TK gene belonged to genotype 1 alongside reference strain Movar33/63.[Conclusions]This study successfully isolated one BoHV-4 strain,providing a reference for further studies on the biological characterization and epidemiology of BoHV-4.关键词
