Abstract

[Objectives]The aim of this study was to evaluate the immune protective effect of candidate antigens for DNA vaccines against Eimeria maxima infection.[Methods]Three vaccine candidate antigens,EmLPL,EmTregIM-1 and EmTregIM-2 from the E.maxima cDNA expression library were screened.Eukaryotic expression plasmids were constructed,namely pVAX1-EmLPL,pVAX1-EmTregIM-1 and pVAX1-EmTregIM-2.Subsequently,nano-DNA vaccines against E.maxima were produced by coating the eukaryotic expression plasmids with PLGA nanoparticles.Finally,the protective efficacies of the nano-DNA vaccines were evaluated through vaccination-challenge experiments.Detection indexes included survival rate,relative weight gain rate,intestinal lesions,oocyst yield and anticoccidial index(ACI).[Results]The results showed that all vaccines significantly reduced body weight loss and oocyst production due to chicken coccidia infection compared to the challenge control group(P<0.05).Except for the pVAX1-EmLPL group,the ACI values of the other five groups ranged from 120 to 160,indicating partial immune protection against E.maxima infection.Among these,the ACI values of the nano-DNA vaccine groups were all above 120 and significantly higher than the corresponding naked plasmid-immunized groups.[Conclusions]Three E.maxima nano-DNA vaccines constructed in this study were all able to provide partial immune protection against E.maxima in chicks.
