摘要

Abstract

In order to induce embryogenic callus of Binglang taro and establish plant regeneration system,the leaf,leaf stalk,thin slice of bulb,stem tip and root of in vitro Binglang taro of Tanbu Wengang in Guangdong province were used as materials to investigate the effects of different plant growth regulators with different concentration on induction,proliferation of embryonic callus and plant regeneration of Binglang taro.The results showed that the best explants for induction of embryogenic callus were the slices on the upper bulb of in vitro taro,with a induction rate of 83.95%.The optimal proliferation medium for embryonic callus was MS+2 mg/L 2,4-D+0.2 mg/L 6-BA,which average proliferation rate was 3.68 times after cultivation of 20 days.MS medium without growth regulator showed the best effect of adventitious bud induction,with differentiation rate of 94.8%,1/2 MS medium supplemented with 0.05 mg/L NAA showed the best effect of rooting,with a rooting rate of 100.00%.The plant regeneration system of embryogenic callus of Binglang taro was established in this study,which provided technical support for the rapid propagation of Binglang taro seedlings,and also provided excellent acceptor materials for the subsequent experiments of polyploid breeding,somatic hybridization and genetic transformation.
