Abstract

A total of 113 898 single nucleotide polymorphism(EST-SNP)markers based on expression sequence tags were successfully identified from the brain transcriptome data of Alosa sapidissima by bioinformatics tools,and 57 polymorphic EST-SNP markers were developed for genetic diversity assessment in A.sapidissima.The minor allele frequency(MAF),the observed heterozygosity(Ho),expected heterozygosity(He)and polymorphism information content(PIC)were 0.067-0.483,0.033-0.567,0.124-0.499 and 0.117-0.375,respectively.Two loci of the 57 EST-SNP significantly deviated from the Hardy-Weinberg equilibrium(P<0.05).The results demonstrated that these polymorphic EST-SNP markers were potent tools for evaluating genetic diversity and germ-plasm analysis of A.sapidissima,which could be capitalized in future conservation strategies and breeding pro-grams to protect and improve this valuable fish.
