Effects of glutaraldehyde and povidone iodine on the immunity and microflora in the hindgut of grass carp(Ctenopharyngodon idella)

Abstract
To investigate the effects of glutaraldehyde and povidone iodine on the immunity and microflora of hind gut in grass carp(Ctenopharyngodon idella),healthy C.idella with average body weight(64.00±0.34 g)were selected and exposed to clear water(control),glutaraldehyde(0.08 mg/L)and povidone iodine(0.05 mg/L),respectively.After one month of feeding,the effects of glutaraldehyde and povidone iodine on the antioxidant en-zyme activities,non-specific immunity and intestinal flora diversity in the hindgut of grass carp were analyzed by enzyme activity,real-time quantitative PCR and high-throughput sequencing.The results showed that glutaralde-hyde and povidone iodine significantly reduced the activity of GPx(P<0.01).The content of MDA significantly increased in the glutaraldehyde group(P<0.01),while mRNA expression levels of immune-related factors(IL-1β,IL-12,TLR4,sIgM,ZO-1,ZO-2,occludin,JAM3,and claudin-1)were significantly lower than the control(P<0.05).The results of microbiome high-throughput sequencing analysis showed that glutaraldehyde did not significantly change the diversity of the hindgut flora of grass carp(P>0.05),however,povidone iodine had a significant effect on the diversity of the hindgut flora of grass carp(P<0.05).Both of these disinfectants affected the proportion of gut dominant flora groups abundance.In conclusion,glutaraldehyde and povidone iodine could reduce the activity of intestinal GPx enzymes,inhibit the intestinal mucosal immune system,reduce immu-nity,and change intestinal flora structure of C.idella,and povidone iodine had a significant effect on the diversity of intestinal microflora.关键词
