Expression and Biological Functions of TRAP1 in Hepatocellular Carcinoma

Abstract
Objective:Tumor necrosis factor receptor-associated protein 1(TRAP1)is a member of the heat shock pro-tein 90 family.However,its role in hepatocellular carcinoma(HCC)has remained unclear.Methods:The expression level and prognosis of TRAP1 in HCC were analyzed based on public databases,followed by the screening of TRAP1 and its related genes to perform gene set enrichment analysis.The effect of silencing TRAP1 on the growth,migration,and intracellular a-denosine triphosphate(ATP)levels were detected in HCC cells.Results:The expression level of TRAP1 significantly in-creased in HCC,and its expression level was related to the grade of HCC(P<0.05).The prognosis of HCC patients in the high TRAP1 expression group was poor(P<0.05).Functional enrichment results showed that TRAP1 and its related genes were associated with gene sets related to gluconeogenesis,mitochondrial functions,the citrate cycle-tricarboxylic acid cycle,and mitochondrial gene modules.Furthermore,TRAP1 knockdown significantly suppressed HCC cell growth,proliferation,and migration,while concurrently reducing intracellular ATP levels.Conclusion:TRAP1 might play a key role in tumorigenesis and progression of HCC by regulating mitochon-drial functions.关键词
