Expression Characteristics of Auxin Transport Carrier Genes in Yam and Their Response to N-(1-Naphthyl)phthalamic Acid

Abstract
In order to investigate the effects of auxin transport carrier genes AUXs and PINs in yam(Di-oscorea opposita)growth development,RT-qPCR technology was used with selected optimal reference genes to study the expression patterns of AUXs and PINs genes in different parts of vines and other tissues,as well as their response to N-(1-naphthyl)phthalamic acid(NPA).The results showed that the stability of eight candi-date reference gens(MYB,GADPH,TRX-2,ACT-1,EF-1α,F-box,HIS,α-TUB)were evaluated using Delta Ct NormFinder,geNorm and Comparative △Ct software,and MYB gene was screened out as the reference gene to analyze the expression of auxin transport carrier genes in different parts of vines.The results of RT-qPCR a-nalysis showed that the DoAUX2,DoAUX3,DoPIN1,DoPIN3 and DoPIN7 genes showed higher expression in the upper part of young yam stems and vines than those in the lower part.The DoAUX1,DoAUX2 and DoPIN3 genes showed higher expression in the upper part of mature vines than those in the lower part.The auxin trans-porter carrier genes were observed in shoot tips,leaves,roots,bulbil,male flowers and female flowers.Among which,the DoAUX1,DoAUX2 and DoPIN7 genes exhibited the highest expression level in leaves,the Do-AUX3 and DoPIN1 genes showed significantly higher expression in shoot tips compared to other tissues,while DoPIN3 and DoPIN6 genes showed higher expression in roots than other tissues,which showed obvious tissue specificity.Compared to the water control,NPA treatment significantly affected the expression of DoAUX1,DoAUX2,DoAUX3,DoPIN1,DoPIN3,DoPIN6,and DoPIN7 genes,and most were down-regulated.In con-clusion,the auxin transport carrier genes regulated the growth and development of yam by regulating the distri-bution of auxin in different tissues,and NPA could affect the transport of auxin in yam plants by regulating the expression of auxin transport carrier genes.关键词
