基于Wnt/β-catenin信号通路探讨固本平喘方对COPD大鼠Wnt1、β-catenin表达的影响
Effect of Guben Pingchuan Formula on the expression of Wnt1 and β-catenin in COPD rats based on Wnt/β-catenin signaling pathway

Abstract
Objective:To investigate the effect of Guben Pingchuan Formula on the expression of Wnt1 and β-catenin in lung tissue of rats with chronic obstructive pulmonary disease(COPD)from the perspective of molecular biology,and to analyze the improvement effect of Guben Pingchuan Formula on COPD-induced airway remodeling.Methods:In this study,64 SPF SD male rats were selected as experimental subjects.According to the random table allocation principle,they were divided into six groups:blank control group,model group,dexamethasone treatment group,and three dose level groups(high dose,medium dose and low dose)of Guben Pingchuan Formula.The rats in the blank control group were fed with adequate food supply and clean environment,and the COPD rats models were established by cigarette smoking and in-tratracheal instillation of porcine pancreatic elastase(PPE 100U/mL)+lipopolysaccharide(LPS 1 mg/ml)in other groups.After the models were successfully established,Guben Pingchuan Formula was administered at a dose of 28 g/(kg·d)in the high dose group,14 g/(kg·d)in the medium dose group,and 7 g/(kg·d)in the low dose group.The dosage of dexametha-sone group was about 0.15 mg/(kg·d).The rats in the model group and the blank control group were given the same amount of normal saline for gavage,and the experiment was carried out continuously for 14 days.HE staining for pathological ex-amination and Western blot were used to detect the expression of Wnt1 and β-catenin in rat lung tissue.Results:Compared with the blank control group,the alveolar wall of rats in the model group was fractured,and the septum was thickened sig-nificantly,accompanying by a large number of inflammatory cell infiltration.In addition,the protein expression levels of Wnt1 and β-catenin were significantly decreased in the model group.Compared with the blank control group,the difference was statistically significant(P<0.05).When compared with the model group,it was found that all dose groups of Guben Pingchuan Formula and dexamethasone group could effectively improve the damage of alveolar structure in rats,especially the dexamethasone group and the high dose group had the most prominent improvement effect.Compared with the model group,the expression of Wnt1 protein in the dexamethasone group and Guben Pingchuan Formula at different doses(low,medium and high)groups was significantly increased(P<0.05).The expression of β-catenin protein in the medium and high dose Guben Pingchuan Formula groups was significantly increased,and the difference was statistically significant.(P<0.05).Conclusion:Guben Pingchuan Formula can improve airway remodeling in COPD rats by regulating the Wnt1 and β-catenin protein expression through the classical Wnt/β-catenin signaling pathway.关键词
