加味芍药甘草汤调控核苷酸结合寡聚化结构域样受体蛋白3炎症小体减轻蛋白尿的实验研究
Experimental study of modified Shaoyao Gancao Decoction on reducing proteinuria by regulating nucleotide-binding oligomerizaed domain-like receptor protein 3 inflammasome

Abstract
Objective:To observe whether modified Shaoyao Gancao Decoction exerts a protective effect on the kid-neys by regulating nucleotide-binding oligomerized domain-like receptor protein 3(NLPR3)inflammasome and reducing proteinuria in IgAN mice.Methods:A BALB/c mouse IgAN model was established,and the mice were randomly divided into a normal control group,a model control group,a modified Shaoyao Gancao Decoction group(2 g/kg),and a valsartan group(12 mg/kg),with 20 mice in each group.From the 12th week,the mice in the normal control group and the model control group were given the same volume of distilled water daily,and the mice in the modified Shaoyao Gancao Decoction group and the valsartan group were given different concentrations of the index drugs,respectively for consecutive 4 weeks.Urine biochemistry was measured,including α1-microglobulin(α1-MG)concentration in urine and serum,β2-microglobu-lin(β2-MG)concentration in urine,N-acetyl-β-D-glucosaminidase(NAG)concentration in urine,and 24-hour urine protein(24 h UP).The levels of blood urea nitrogen(BUN)and Serum creatinine(Scr)were measured in mice.HE staining was used to observe the pathological changes in mice kidney tissue.The expression levels of NLRP3 antibody were detected by immunohistochemistry.Results:Compared with those of the model control group,the levels of urine biochemistry(in-cluding urine α1-MG concentration,urine β2-MG concentration,urine NAG concentration and total 24 h UP),serum BUN,Scr concentrations,and NLPR3 inflammasome protein expression level in the valsartan group and modified Shaoyao Gancao Decoction group were significantly decreased,with statistical significance(P<0.05).Compared with those of the valsartan group,the urine biochemical levels(including urine α1-MG concentration,urine β2-MG concentration,urine NAG concentration and total 24 h UP),serum BUN,Scr concentration and NLPR3 inflammasome protein expression level in modified Shaoyao Gancao Decoction group were significantly decreased,with statistical significance(P<0.05).Conclu-sion:modified Shaoyao Gancao Decoction has a good therapeutic effect on IgAN mice,which can regulate NLPR3 inflam-masome,inhibit inflammatory response,and alleviate kidney damage in IgAN mice.关键词
