Effect of electroacupuncture on the expression of NR2B and PP2A in the anterior cingulate cortex of inflammatory pain mice

Abstract

Objective To observe the effects of electroacupuncture(EA)on the behaviors of mice with complete Freund's adjuvant(CFA)-induced inflammatory pain and the expression of N-methyl-D-aspartate receptor subunit 2B(NR2B)and protein phosphatase 2A(PP2A)in the anterior cingulate cortex(ACC),and to discuss the role of NR2B and PP2A in the analgesic effect of EA.Method Twenty-four male C57BL/6 mice were randomly allocated to a control group,a model group,and an EA group,each consisting of 8 mice.Mice in the model group received an injection of 25 μL of CFA at the right hind paw to establish inflammatory pain models.Mice in the EA group received EA at Zusanli(ST36)and Sanyinjiao(SP6)on the right side at post-modeling day 1,day 3,day 5,and day 7.The mechanical paw withdrawal rate and thermal paw withdrawal latency of both hind paws were observed in the three groups of mice.The protein expression of NR2B and PP2A in the ACC was detected using the Western blot(WB),immunohistochemistry(IHC),and immunofluorescence(IF)assays.Result Compared to the control group,the mechanical paw withdrawal rate increased notably(P<0.05)and the thermal paw withdrawal latency dropped significantly(P<0.05)in the model group at post-modeling day 1,day 3,day 5,and day 7.Compared to the model group,the mechanical paw withdrawal rate decreased notably(P<0.05)and the thermal paw withdrawal latency increased markedly(P<0.05)in the EA group at post-modeling day 1,day 3,day 5,and day 7.The WB results demonstrated that the model group had decreased PP2A protein expression(P<0.05)and increased NR2B protein expression(P<0.05)in the ACC compared to the control group;compared to the model group,the expression of PP2A increased notably(P<0.05)and the expression of NR2B dropped significantly(P<0.05)in the EA group.The IHC findings showed that compared to the control group,the model group had notably decreased PP2A expression in the ACC(P<0.05),and the EA group had increased PP2A expression compared to the model group(P<0.05).The IF assay revealed that compared to the control group,the model group had significantly increased NR2B protein expression in the ACC(P<0.05),and the EA group had decreased NR2B protein expression compared to the model group(P<0.05).Conclusion EA can improve hyperalgesia in inflammatory pain mice by down-regulating the expression of NR2B and up-regulating the expression of PP2A in the ACC,indicating that NR2B and PP2A may play a role in EA's effects in reducing inflammatory pain.
