摘要

Abstract

This study optimized the extraction process of polyphenols from eucalyptus leaves using deep eutectic solvents with choline chloride and betaine as hydrogen bond acceptors through single-factor and orthogonal experiments,and evaluated their antioxidant activity.The results showed that the optimal conditions were as follows:A betaine-to-ethylene glycol ratio of 2∶1,water content of 50%,liquid-to-material ratio of 40 mL/g,and ultrasonic power of 300 W,ultrasonic temperature of 75℃,ultrasonic time of 50 min.Under these conditions,the extraction yield of eucalyptus polyphenols reached 22.39%,significantly higher than that of traditional solvents.The eucalyptus leaf extract exhibited stronger scavenging effects on 2,2-diazo-di(3-ethyl-benzothiazolin-6-sulfonic acid)diamiammonium salt(ABTS+)radicals compared to 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)and nitrogen dioxide radicals,with maximum scavenging rates of 98.8%,83.5%,and 77.5%,respectively.The corresponding IC50 values were 4.41,4.82,and 324.70 mg/L.The study demonstrates that the betaine/ethylene glycol-based deep eutectic solvents system achieves high extraction efficiency and excellent antioxidant activity for eucalyptus polyphenols,providing a reference for the future development and utilization of eucalyptus resources.
