Abstract

The study aimed to optimize the microwave-assisted deep eutectic solvent(DES)extraction process of Bombax malabaricum DC.polysaccharides and evaluate the antioxidant activity of the polysaccharides before and after purification.The DES composed of choline chloride and formic acid at a molar ratio of 1∶1 was selected as the extraction solvent.Based on single-factor experiments,the water content of DES,liquid-to-solid ratio,microwave temperature,and extraction time were chosen as independent variables,while the extraction yield of Bombax malabaricum DC.polysaccharides was the dependent variable,response surface methodology was employed to optimize the extraction process.Using vitamin C(VC)as a positive control,the scavenging abilities of Bombax malabaricum DC.polysaccharides before and after purification against 1,1-diphenyl-2-picrylhydrazyl(DPPH)radicals and hydroxyl radicals were evaluated.The results showed that the optimal extraction conditions were:DES water content of 42%,liquid-to-solid ratio of 40 mL/g,microwave temperature of 108℃,and extraction time of 21 min.Under these conditions,the extraction yield of Bombax malabaricum DC.polysaccharides reached 174.9 mg/g.The antioxidant activity tests showed that the scavenging ability of Bombax malabaricum DC.polysaccharide on DPPH and hydroxyl free radical increased with the increase of mass concentration of 0.1~3.0 g/L,and the scavenging ability of purified Bombax malabaricum DC.polysaccharides on DPPH and hydroxyl free radical was significantly stronger than that of Bombax malabaricum DC.polysaccharides.The study indicates that microwave-assisted DES extraction of Bombax malabaricum DC.polysaccharides is green and efficient,providing a reference for its application in feed research.
