Abstract

Gymnocypris przewalskii,found only in Qinghai Lake and its tributaries,is the only Cyprini-dae species adapted to the extreme ecological environment of Qinghai Lake.The release of artificially bred G.przewalskii to Qinghai Lake has been carried out for more than 20 years,but the effects on genetic diversity and population structure are not clear.In this study,G.przewalskiispecimens were collected over three years(2020-2022)from four populations:artificially bred juveniles(FL),wild(SS),and conserved(BZ),with the prerelease historical population(LS)serving as the reference.The phenotypic traits,genetic diversities and population structures of the four G.przewalskii populations were compared using morphological characteristics and Cyt b gene markers.Our objectives were to evaluate effects of the large-scale release of artificially bred G.przewalskii on the population in Qinghai Lake and provide a scientific basis for implementing more effective conservation measures.Morphological characteristics between the SS and LS populations,such as outer gill raker number and the ratio of body length to body depth,were not significantly different(P>0.05).A total of 3 461 cytochrome b(Cyt b)sequences of length 1 140 bp were obtained for the four populations,and each population produced 20 variant loci and 23 hap-lotypes.The haplotype diversity index and nucleotide diversity index of the four populations were in the range of 0.762-0.846 and 0.00177-0.00204,respectively,and there were no significant differences among the populations(P>0.05).A constructed haplotype network showed that the genetic structure of the four G.przewalskii populations was identical.In conclusion,the genetic diversity and population structure of G.przewalskii in Qinghai Lake was well maintained,with little or no influence by artificial breeding and release.
