Abstract

Objective To explore the effect of arsenate methyltransferase(AS3MT)on the viability and apoptosis of human hepatocel-lular carcinoma cells(HepG2)exposed to arsenic.Methods Using public databases such as TCGA and GTEx,the expression levels of AS3MT in normal tissues and tumor tissues of patients with different cancers were analyzed,and its correlation with DNA methyla-tion sites and immune cell infiltration in LIHC patients,and its prognostic value were further analyzed.HepG2 cells were exposed to 0,8,16,24 μmol/L NaAsO2,and then AS3MT expression was quantitatively analyzed using RT-qPCR.HepG2 cells were transfected with siRNAs and subsequently exposed to arsenic,named as NC group,siAS3MT#1 group,siAS3MT#2 group,siAS3MT#1+24 μmol/L NaAsO2 group,siAS3MT#2+24 μmol/L NaAsO2 group,respectively.The transfection efficiency of AS3MT was detected using RT-qPCR,and cell viability and apoptosis of HepG2 cells were evaluated using MTT assay,Hoechst 33342 staining,and propidium io-dide(HO/PI)staining.Results Database analysis results showed that AS3MT expression exhibited significant differences between 27 types of tumor tissues and normal tissues(P<0.05),and the expression of AS3MT was significantly higher in LIHC than in normal tissue(t=0.578,P=0.007).Furthermore,AS3MT expression was significantly correlated with multiple DNA methylation sites in LIHC patients(P<0.05).Survival analysis revealed that the overall survival rate of LIHC patients in AS3MT low expression group was signifi-cantly higher than that in high expression group(HR=0.63,P=0.009).Additionally,AS3MT expression was an independent prognostic factor for LIHC patients(P=0.009).Moreover,the expression of AS3MT was correlated with the infiltration of multiple immune cells(P<0.05).Compared with 0 μmol/L NaAsO2,the expression level of AS3MT in HepG2 exposed to 16,24 μmol/L NaAsO2 was signifi-cantly increased(P<0.01).Compared with NC group,the cell viability was decreased in siAS3MT#1 group and siAS3MT#2 group(P<0.05),and the apoptosis was increased.Compared with the corresponding siAS3MT group,the viability of HepG2 cell was further decreased in siAS3MT+24 μmol/L NaAsO2 group(P<0.01),and the apoptosis was increased.Conclusion AS3MT exhibits high expres-sion in arsenic-exposed HepG2 cells.Specific silencing of AS3MT significantly inhibits the viability of arsenic-exposed HepG2 cells and induces apoptosis,suggesting that AS3MT has potential as a biomarker and therapeutic target for hepatocellular carcinoma.
