DHX32对胃癌细胞增殖和凋亡的影响
Effect of DHX32 on cell proliferation and apoptosis of gastric cancer

Abstract
Objective To study the effect of DHX32 on the cell proliferation and apoptosis of gastric cancer.Methods From July 2021 to May 2022,36 cases of gastric cancer and para-cancer tissue samples in the department of pathology of Shaanxi Provincial Tu-mor Hospital were collected.RNA and protein were extracted from tissue samples to detect DHX32 expression by qRT-PCR and West-ern blot.Small interfering RNAs of DHX32 were trasfected to MKN-28 and AGS cells to silence the expression of DHX32.Gastric can-cer cells were divided into negative control group(invalid siRNA),DHX32 siRNA-1 group and DHX32 siRNA-2 group.Then cell pro-liferation ability was detected by MTT method,and cell cycle and cell apoptosis were detected by flow cytometer sorting.DHX32 and β-catenin protein expression levels were examined by Western blot.Results Quntitative RT-PCR results showed that DHX32 mRNA expression was significantly higher in gastric cancer tissues than in para-cancer tissues(P<0.01).Western blot results showed DHX32 protein expression level was also higher in gastric cancer tissues than in para-cancer tissues(P<0.01).After DHX32 was silenced in gastric cancer cells,the cell proliferation activity in siRNA-1 and siRNA-2 groups were significantly reduced compared with negative control group(P<0.01).Compared with negative control group,G0/G1 phase cell percentage was enhanced in DHX32 siRNA-1 and siRNA-2 groups(P<0.01),and percentages of S phase and G2/M phase cells were significantly reduced(P<0.01).The percentages of early and late apoptotic cells in DHX32 siRNA-1 and siRNA-2 groups were significantly increased compared with negative control group(P<0.01).DHX32 and β-catenin protein expressions were significantly downregulated in siRNA-1 and siRNA-2 groups com-pared with negative control group(P<0.01).Conclusion DHX32 expression is significantly upregulated in gastric cancer tissues,and it can promote the cell proliferation and inhibit the apoptosis of gastric caner cell by upregulating β-catenin expression.关键词
