miR-215-5p在肾透明细胞癌中的表达及其对786-O细胞功能的影响
Expression of miR-215-5p in clear cell renal cell carcinoma and its effects on biological functions of 786-O cells
摘要
Abstract
Objective To investigate the expression of miR-215-5p in clear cell renal cell carcinoma(ccRCC)and its effects on the functions of 786-O cells.Methods The expression profile of miR-215-5p in ccRCC was analyzed using The Cancer Genome Atlas(TCGA)database.Real-time quantitative polymerase chain reaction(RT-qPCR)was performed to detect miR-215-5p expression levels in 293T,786-O,and ACHN cells.786-O cells were respectively transfected by transient transfection technology,named as inhibitor NC group and miR-215-5p inhibitor group.The transfection efficiency was validated by RT-qPCR.Cell migration ability was as-sessed using scratch wound healing assay,cell invasion capability was evaluated through Transwell chambers,cell proliferation was measured by CCK-8 assay,and epithelial-mesenchymal transition(EMT)-related protein expression was detected by Western blot.Results TCGA data revealed that miR-215-5p expression was significantly higher in ccRCC tissues than in adjacent normal tissues(P<0.05),and it was higher in male than in female(P<0.05).RT-qPCR confirmed that miR-215-5p expression was higher in ccRCC cells than in normal renal cells(P<0.05).Compared with inhibitor NC group,the abilities of cell migration,cell proliferation,and invasion were decreased in miR-215-5p inhibitor group(P<0.01),and Vimentin protein expression was downregulated(P<0.05).Conclusion miR-215-5p is highly expressed in ccRCC cells,and its knockdown suppresses proliferation,migration,invasion,and EMT progres-sion,which provides a new potential target for the treatment of ccRCC.关键词
