Piezo1 promotes invasion and migration of cervical cancer cells by activating YAP signaling pathway

Abstract
Objective To investigate the expression of Piezo1 in cervical cancer and its role in the invasion and migration of cervical cancer,and explore the related molecular mechanism.Methods A total of 20 cervical cancer tissues and 15 normal cervical tissues from patients who underwent total hysterectomy due to uterine fibroids or adenomyosis were collected.The expression of Piezo1 protein in cervical cancer tissues and normal cervical tissues was detected by immunohistochemistry.The cervical cancer cell lines HeLa and SiHa were divided into six groups:control group,sh-NC group,sh-Piezo1 group,Yoda1(Piezo1 agonist)group,Verteporfin(YAP-TEAD complex inhibitor)group,and Yoda1+Verteporfin group.Cell migration was detected by scratch assay,cell invasion was de-tected by Transwell assay,Piezo1 gene expression level was detected by qRT-PCR,and the protein expression levels of Piezo1,YAP,and epithelial-mesenchymal transition(EMT)markers E-cadherin and Vimentin were detected by Western blot.Results Compared with normal cervical tissues,Piezo1 protein was highly expressed in cervical cancer tissues(P<0.05).Compared with sh-NC group,the mRNA and protein expression levels of Piezo1 were significantly reduced in sh-Piezo1 group(P<0.05),the relative migration rate and the invasion number were decreased(P<0.05),E-cadherin expression was increased while Vimentin expression was decreased(P<0.05),and the nuclear localization of YAP protein was reduced.Compared with control group,the relative migration rate and the number of invasions were increased in Yoda1 group(P<0.05),the expression of E-cadherin was decreased while the expression of Vimentin was increased(P<0.05),and the nuclear localization of YAP was enhanced.Compared with control group,the relative migration rate,the number of invasion,and EMT progression were decreased in Verteporfin group(P<0.05).Compared with Yoda1 group,the rela-tive migration rate,the number of invasion,and EMT progression were decreased in Yoda1+Verteporfin group(P<0.05).Conclusion Piezo1 is highly expressed in cervical cancer.Piezo1 may promote the migration,invasion,and EMT of cervical cancer cells by activat-ing YAP signaling pathway.关键词
