细胞磁力加载平台对牙周膜干细胞排列和胶原分泌的影响
Effect of cell magnetic loading platform on arrangement and collagen secretion of periodontal ligament stem cells

Abstract
Objective To construct a cell magnetic loading platform for simulating the mechanical microenvironment of periodontal ligament stem cells(PDLSCs)and investigate its influence on the arrangement and collagen secretion of PDLSCs.Methods Both ends of the rat tail collagen hydrogel scaffold were modified by magnetic micron-sized particles,and then given 10%uniaxial static stretching by magnetic force to construct a cell magnetic loading platform.The morphology of collagen scaffold was observed under scanning electron microscope(SEM)and laser confocal microscope reflection mode(CRM).PDLSCs were then cultured on the surface of the collagen scaffold,and divided into three groups:control group,pre-stretching for 36 h group and continuous magnetic stretch-ing group.Cytoskeleton staining was used to observe the orientation of PDLSCs.Immunofluorescence was used to observe the arrange-ment direction of type Ⅰcollagen in each group.The arrangement angles of cell and human type Ⅰ collagen was calculated to compare the differences in the direction of PDLSCs and type Ⅰ collagen among all groups.Results SEM and CRM results showed that the fi-bers of the collagen scaffold aligned along the stretching direction after 10%uniaxial static magnetic stretching.The direction of PDLSCs and secreted type Ⅰ collagen in pre-stretching for 36 h group and continuous magnetic stretching group tended to be consis-tent with the direction of magnetic stretching.Compared with control group and pre-stretching for 36 h group,the alignment angles of PDLSCs and secreted type Ⅰ collagen in continuous magnetic stretching group were more tended to 0°.Conclusion The cell mag-netic loading platform is successfully constructed by 10%uniaxial static magnetic stretching on the collagen hydrogel scaffold.Under the action of static magnetic stretching,the orientation of PDLSCs and secreted type Ⅰ collagen is all tended to the direction of mag-netic stretching,providing a research basis for the orderly regeneration of periodontal ligament fibers.关键词
