Abstract
Objective To investigate the effect of recombinant humanized type Ⅲ collagen(rhCol Ⅲ)solution on cytotoxicity and ad-hesion of human corneal epithelial cells,and explore its ameliorating effect on benzalkonium chloride-induced dry eye syndrome.Methods Human corneal epithelial cells were divided into negative control group,positive control group,rhCol Ⅲ group,rhCol Ⅲsolution group,moisturizing solution group and sodium hyaluronate group.After 24 h intervention,CCK-8 was used to detect cell vi-ability.After partial removal of the third eyelid,14 Japanese white rabbits were randomly divided into five groups:normal group,model group,rhCol Ⅲ solution group,moisturizing eye drops group and negative control group.The dry eye syndrome model was in-duced by instilling 0.2%benzalkonium chloride drops.Subsequently,the corresponding drugs were consecutively administered for 14 d.The phenol red thread test was used to measure the amount of tear secretion.Corneal fluorescein sodium staining was utilized to assess the area of corneal damage.The tear ferning test was applied to examine tear crystallization.Hematoxylin-eosin staining was used to ob-serve the pathological changes in the cornea.Results Compared with negative control group,the viability of human corneal epithe-lial cells showed no significant difference in rhCol Ⅲ group,rhCol Ⅲ solution group,moisturizing solution group and sodium hyaluro-nate group,while the cell viability and the adhesion capacity were enhanced in rhCol Ⅲ group and rhCol Ⅲ solution group(P<0.01).After 14 d treatment,compared with normal group,the tear secretion was significantly increased in both rhCol Ⅲ solution group and moisturizing eye drops group(P<0.01),and the corneal injury area was decreased(P<0.01).After 14 d treatment,the tear ferning crystallization exhibited small branches and incomplete morphology in moisturizing eye drops group while the crystal density was en-hanced with well-defined morphological integrity in rhCol Ⅲ solution group.The recovery state of the corneal conjunctival epithelium in rhCol Ⅲ solution group was closer to that of normal group than in moisturizing solution group.Conclusion Recombinant humani-zed type Ⅲ collagen solution can enhance the cell viability and the adhesion capacity of human corneal epithelial cells.It can also ef-fectively alleviate the symptoms of dry eye syndrome by stimulating the tear secretion in rabbits,repairing the corneal damage,and in-ducing the repair and healing of corneal and conjunctival epithelia.关键词
