抗苗勒管激素诱导多囊卵巢综合征样小鼠表型

Abstract
Objective:To establish a mouse model of polycystic ovary syndrome(PCOS)using anti-Müllerian hormone(AMH). Methods:By a random number table method,C57BL/6 female mice aged 21 days were divided into four groups:the control group without treatment(n=5),dehydroepiandrosterone(DHEA)group with daily subcutaneous injection of DHEA(60 mg·kg-1·d-1)for 21 days(n=5),LAMH group with intraperitoneal injection of AMH(0.5 mg/kg)once a week for 4 weeks(n=5),H-AMH group with intraperitoneal injection of AMH(0.5 mg/kg)twice a week for 4 weeks(n=5).The mice were weighed weekly.The DHEA group received vaginal smear at the 4th and 5th week of modeling.In the AMH group with different doses,estrous cycles were observed by vaginal smear at the 5th and 6th week of modeling.Sex hormone levels of each group were detected by enzyme-linked immunosorbent assay(ELISA).The histological changes of the ovary were observed by hematoxylin and eosin(HE)staining. Results:After the intervention,the body weight of mice in the DHEA group was significantly higher than those in other groups(P<0.01).The estrous cycles of mice in the DHEA group,the L-AMH group and the H-AMH group were stagnant after modeling,and the DHEA group as well as the H-AMH group mainly showed irregular and incomplete estrous cycle.HE staining results showed that the ovarian tissue of mice in the DHEA group and the H-AMH group showed obviously polycystic changes.The detection of serum sex hormone levels found that testosterone(T),LH in DHEA and H-AMH groups were significantly higher and the levels of FSH and estradiol(E2)were significantly lower than those in the control group(P<0.05).Compared with the DHEA group,serum T and LH levels were significantly decreased,while serum FSH and E2 levels were significantly increased in L-AMH and H-AMH groups(all P<0.05).Compared with the H-AMH group,serum T level was significantly decreased,serum FSH and E2 levels were significantly increased in the L-AMH group(all P<0.001). Conclusions:Compared with the control group,similar pathological changes were found in the H-AMH group and the DHEA group,and both abnormal reproductive and metabolic changes were similar to PCOS patients.关键词
