Abstract

Lignocellulose is the most abundant renewable resource in nature,and its biorefin-ery requires the use of cellulase for hydrolysis,but the low yield and high production costs significantly limit the large-scale industrial application of cellulase.To obtain high cellulase-producing bacterial strains,a cellulase-producing strain LT-4 was isolated and purified from the soil of sugarcane land in Liuzhou,Guangxi,by using Congo red-carboxymethyl cellulose sodium medium.The strain was then identified through molecular biology techniques,and the fermentation conditions for cellulase production were optimized using single-factor and or-thogonal experiments.The results showed that the strain LT-4 belongs to the genus Bacillus sp.Using Avicelase as the evaluation index,the optimal liquid fermentation conditions for cellulase production were as follows:Using a carbon source of wheat bran and sugarcane ba-gasse in a mass ratio of 4∶1,4 g/L of peptone as the nitrogen source,a fermentation broth pH of 8.0,and incubation at 39 ℃ for 36 h.After optimization,the optimized Avicelase activ-ity reached(0.237±0.014)U/mL,which is 2.3 times higher than that before optimiza-tion.This study is of great significance for improving cellulase yield and reducing production costs,providing a feasible solution for the efficient utilization and resource conversion of lignocellulose,while also laying a foundation for industrial applications in related fields.
