苦参碱衍生物体外抑制白色念珠菌生长及其作用机制
Inhibition of the growth of Candida albicans in vitro by matrine derivatives and its mechanism of action

Abstract
Candida albicans is a common human pathogen that can cause candida vaginitis.Candida vaginitis is a common mucosal infection of the female reproductive tract.The com-mon symptoms include vaginal itching,pain,redness,etc.At the same time,the recurrence rate is high,which causes serious distress to female patients.In the present study,a series of experiments were carried out to investigate the inhibitory effect of matrine and its derivative a~h on Candida albicans and its mechanism of action.The MIC of matrine was 5 mg/mL,and the MIC of matrine derivative c was 0.625 mg/mL.After derivatization,the antibacterial activity of matrine derivatives was significantly better than that of matrine.By monitoring the growth of Candida albicans,it can be obtained that derivative c can inhibit the growth of Candida albicans and prolong its slow growth period.The results of field emission scanning electron microscopy showed that the Candida albicans after the treatment of derivative c had broken,deformed and depressed on its surface.By detecting the leakage of cellular proteins,the membrane permeability of Candida albicans was increased after matrine derivative c treatment.By propidium iodide staining,N-phenyl-1-naphthylamine staining and β-galactosi-dase assay,it was further verified that matrine derivative c could destroy the cell membrane of Candida albicans,increase the permeability of cell membrane,and cause the death of Can-dida albicans.Rhodamine 123 staining was used to explore the changes in the membrane po-tential of Candida albicans under the action of matrine derivative c,and it was found that matrine derivative c could destroy the membrane potential of Candida albicans,thereby af-fecting the normal physiological activities of bacteria and eventually causing the death of Candida albicans.The level of reactive oxygen species(ROS)in the cytoplasm of Candida albicans was determined using 2,7-dichlorofluorescein diacetate,and the results showed that the oxidation-reduction balance of Candida albicans was disrupted by the alkaloid derivative c of matrine,leading to a large accumulation of ROS and cell death.Through a series of ex-periments,it was found that matrine and its derivatives can inhibit the growth of Candida al-bicans by disrupting the structure of the cell membrane and increasing its permeability,provi-ding a new idea for the mechanism of matrine inhibiting Candida albicans.It is expected to provide new ideas and strategies for the treatment of candidal vaginitis.关键词
