解毒活血汤对慢性肾脏病大鼠TGF-β1、α-SMA表达的影响
Effect of decoction for removing toxin and activating blood on expressions of TGF-β1 and α-SMA in rats with chronic kidney disease

Abstract
Objective It is to investigate the effect of decoction for removing toxin and activating blood on chronic kid-ney disease(CKD)in rats via transforming growth factor-β(TGF-β).Methods Seventy-two SPF-grade male SD rats were randomly divided into blank group,model group,Niao-Du-Qing group,high dose,medium dose and low dose groups of de-coction for removing toxin and activating blood,with 12 rats in each group.The rats of the blank group were not treated,while the rats in the other groups were given adenosine and high-phosphorus diet to establish models of CKD.From the 5th to the 8th week of the experiment,the Niao-Du-Qing group was given Niao-Du-Qing granules 0.025 μg/(kg·d)by ga-vage,the high dose,medium dose and low dose groups of decoction for removing toxin and activating blood were given de-coction for removing toxin and activating blood 5.6 g/(kg·d),2.8 g/(kg·d),and 1.4 g/(kg·d)by gavage,respec-tively,the blank group and model group were given an equal amount of 0.9%NaCl by gavage,all once daily.The levels of calcium(Ca),phosphorus(P),blood urea nitrogen(BUN)and creatinine(SCr)in serum were determined by bio-chemical assay kits,the histopathological morphology of renal tissues were observed by HE staining,and the mRNA expres-sions of α-smooth muscle actin(α-SMA)and TGF-β1 in renal tissue were determined by Real-time PCR.Results Com-pared with the blank group,the levels of SCr,P,and BUN in rats of the model group were significantly increased(all P<0.05),while the level of blood calcium was significantly decreased(P<0.05);collagen deposition and fibrous streak for-mation,renal interstitial edema,and inflammatory cell infiltration were found in renal tissues;the relative mRNA expres-sions of TGF-β1 and α-SMA in renal tissues were significantly increased(both P<0.05).Compared with the model group,the levels of blood P,SCr and BUN in rats of the Niao-Du-Qing group,high dose,medium dose groups of decoction for removing toxin and activating blood were significantly reduced(all P<0.05),and the levels of blood calcium were sig-nificantly increased(all P<0.05);the structural damage of the kidneys and renal fibrosis were alleviated to different de-grees;the relative mRNA expressions of TGF-β1 and α-SMA in renal tissues were significantly reduced(all P<0.05).The levels of SCr,P,and BUN in rats of the Niao-Du-Qing group and high dose group of decoction for removing toxin and activating blood were significantly lower than those in the medium dose and low dose groups of decoction for removing toxin and activating blood(all P<0.05),and the levels of blood calcium were significantly higher than that in the low dose group of decoction for removing toxin and activating blood(P<0.05);the improvement of lesions in renal tissues was more significant;the relative mRNA expressions of TGF-β1 and α-SMA in renal tissues were significantly lower than those in the medium dose and low dose groups of decoction for removing toxin and activating blood(all P<0.05).Conclusion Decoc-tion for removing toxin and activating blood can improve renal function and alleviate lesions of renal tissues of rats with chro-nic kidney disease,the mechanism may be related to regulate TGF-β pathway.关键词
